Ribosome-inactivating proteins (RIPs) are rRNA N-glycosylases isolated mainly from plants that catalyze the hydrolysis of the N-glycosidic bond of a specific adenosine in the sarcin-ricin loop (SRL) of the major ribosomal RNA. Because the SRL is crucial for anchoring translation elongation factors, RIPs cause inactivation of ribosomes. They have been classified into two types based on the presence (type 2 RIPs) or absence (type 1 RIPs) of a lectin chain that can turn type 2 RIPs into potent toxins, such as ricin or abrin. The biological role of these proteins is unknown, but they are thought to be a defense mechanism of some plants against pathogens and predators. Because of their enzymatic action, RIPs show several biological activities, among which antiviral, antifungal and antiproliferative activities stand out. The most promising application of RIPs is their use as a component of immunotoxins, in which RIPs are linked to antibodies that mediate their binding and internalization by malignant cells. In agriculture, RIPs have been shown to increase resistance against viruses, fungi and insects in transgenic plants. The studies collected in this book provide the reader with an overview of the most current and interesting lines of research in the field of RIPs and their applications in medicine and agriculture. Thus, the reprint includes the isolation and biological properties of some new RIPs, both type 1 and type 2, the mechanisms of toxicity of



previously described RIPs and two extensive reviews, one on the antiviral activity of RIPs and the other on the strategies used to improve their pharmacological properties.

Malignant mesothelioma (MM) is a rare and aggressive cancer, related to chronic inflammation and oxidative stress caused mainly by exposure to asbestos. Although this mineral has been banned for decades in many countries, epidemiologists predict the MM epidemic will last past 2040, raising many concerns in public health given its late diagnosis, dismal prognosis, and lack of current efficient therapies.To deal with this situation, important breakthroughs have recently been made in the understanding of MM's complex biology and the carcinogenic process of the different patterns of the disease. Examples of these include the development of new biomarkers and the deciphering of gene-environment interactions, molecular mechanisms of invasiveness, deregulated pathways, altered expression of miRNAs, DNA damage repair, or metabolic profile. From now on, MM's aggressive and chemoresistant character appears linked to a polyclonal malignancy, and heterogeneity in molecular alterations.Given these improvements, new therapeutic targets are being explored to solve the double challenge faced by clinicians. The first is to reduce tumor development and its wasting consequences as soon as possible,



without resistance and with limited toxicity. The second is to stimulate the recognition of tumor cells by the induction of a specific immune response. This Special Issue will highlight all these aspects.

Sommario/riassunto

This book aims at an innovative approach within the framework of convex analysis and optimization, based on an in-depth study of the behavior and properties of the supremum of families of convex functions. It presents an original and systematic treatment of convex analysis, covering standard results and improved calculus rules in subdifferential analysis. The tools supplied in the text allow a direct approach to the mathematical foundations of convex optimization, in particular to optimality and duality theory. Other applications in the book concern convexification processes in optimization, non-convex integration of the Fenchel subdifferential, variational characterizations of convexity, and the study of Chebychev sets. At the same time, the underlying geometrical meaning of all the involved concepts and operations is highlighted and duly emphasized. A notable feature of the book is its unifying methodology, as well as the novelty of providing an alternative or complementary view to the traditional one in which the discipline is presented to students and researchers. This textbook can be used for courses on optimization, convex and variational analysis, addressed to graduate and post-graduate students of mathematics, and also students of economics and engineering. It is also oriented to provide specific background for courses on optimal control, data science, operations research, economics (game theory), etc. The book represents a challenging and motivating development for those experts in functional analysis, convex geometry, and any kind of researchers who may be interested in applications of their work.
